I want to replace the crankcase gasket, and inspect the main bearings.In order to remove the cylinder from it's base, do I have to disconnect the connecting rod through the port holes first, or can I remove the cylinder and crankshaft from the base as one unit? Are the main bearings one piece, or two? Are they adjustable? Thanks.

It should be split bearings, if there's no play in them I wouldn't worry about them.
